Resolving dynamic OGT-protein interactions reveals its use of adaptor proteins to control cell state transitions
The sole O-GlcNAc transferase, OGT, interacts with nearly 1,000 proteins and glycosylates thousands of proteins in a site-specific manner. How OGT targets its substrates is poorly understood, and the role of the resultant glycosylation events are largely uncharacterized. Here, we demonstrate that OGT uses specific proteins and/or protein complexes to target downstream substrates, which in turn, controls some of the earliest decisions in the developing zygote. We will also discuss technical challenges to study dynamic protein-protein interactions in cells with differing background proteomes, and provide several solutions to better classify false positives and rescue false negatives.
